检测项目
定性分析:通过比较未知样品与标准物质的紫外吸收光谱特征峰位置和形状,对化合物的结构类型或官能团进行初步识别和确认。
定量分析:依据朗伯-比尔定律,测量特定波长下样品的吸光度值,建立标准曲线,从而精确测定样品中特定组分的浓度含量。
纯度检查:利用紫外光谱检测样品中是否存在杂质吸收峰,或通过吸收度的比值判断原料药或化学试剂的纯度是否符合规定要求。
异构体鉴别:根据不同几何异构体或立体异构体在紫外区的吸收特性差异,对顺反异构体或互变异构体等进行区分和鉴定。
氢键强度测定:通过观察极性溶剂与非极性溶剂中化合物紫外吸收峰的位移现象,分析和评估分子内或分子间氢键的强度及形成情况。
反应动力学研究:监测化学反应过程中反应物或产物特征吸收峰强度随时间的变化规律,推算反应速率常数和反应机理。
络合物组成测定:利用等摩尔连续变化法或摩尔比法,根据络合物形成前后紫外吸收光谱的变化,确定络合物的组成比和稳定常数。
DNA/蛋白质浓度测定:基于核酸在260纳米附近或蛋白质在280纳米附近的特征吸收,快速测定生物样本中DNA或蛋白质的浓度。
